Overview (short): 5-Amino-1MQ (also written 5-amino-1-methylquinolinium / 5-AMQ / 5-Amino-1MQ) is a small-molecule inhibitor developed to block nicotinamide N-methyltransferase (NNMT). It has been used widely in preclinical research to probe NNMT’s role in metabolism, adiposity, NAD⁺ biology, and cancer cell energetics. Mechanism of action — NNMT inhibition and cellular metabolism NNMT is an enzyme that methylates nicotinamide (NAM) to form 1-methyl nicotinamide (MNA), consuming the methyl donor S-adenosylmethionine (SAM) and altering cellular NAD⁺ salvage dynamics.
